Mechanism of the biological effect of the tumor necrosis factor-аlpha at psoriasis

Abstract

Among different cytokines, tumor necrosis factor-аlpha (tumor necrosis factor α, TNF-α) plays a special role in psoriatic immunopathogenesis. Data on this cytokine collected for the recent decades made it possible to create a number of biological drugs blocking TNF-α, which are successfully applied in clinical practice for treating medium to severe psoriasis and psoriatic arthritis. This review presents general information about the cytokine structure and its receptor apparatus, regulation mechanisms of TNF-α synthesis and ways of signal transmission as the basis needed to implement the biological effects of cytokine in the development of psoriatic skin affections.

Functions of Toll-like receptors as an inborn immunity component and their participation in the pathogenesis of dermatoses of different etiologies

Abstract

In addition to serving as a mechanical barrier protecting our organism from the damaging effect of different factors, our skin also takes part in immune reactions developing in case of microbial intervention. Toll-like receptors (TLR) mediating recognition of molecular structures of pathogens are expressed in skin cells of different types initiating the development of adaptive immune reactions when associated with different ligands. The review presents data on the structure and functions of TLR, their localization in skin compartments and their role in the pathogenesis of skin diseases being of infectious etiology and non-infectious origin.

Role of polymorphism of the androgen receptor gene and non-random X chromosome inactivation in the genesis of androgenic alopecia in women of childbearing potential

Abstract

The authors describe the results of a study of polymorphism of the androgen receptor gene by the number of CAG repeats in exon 1 of the androgen receptor gene and non-random X chromosome inactivation in 87 women of childbearing potential (at the average age of 29.5 ± 5.4 years) suffering from androgenic alopecia. They revealed an association between the presence of 'short' (≤ 22) CAG repeats in both alleles of the androgen receptor gene (р < 0,05) and a reliable growth of prevalence of non-random X chromosome inactivation in patients with androgenic alopecia as compared to healthy women (50.7% (39/77) and 16.1% (9/56), respectively, р < 0.05). These data demonstrate a pathogenetic role of polymorphism of the androgen receptor gene and non-random X chromosome inactivation in the development of androgenic alopecia in women of childbearing potential as well as urgency of using molecular and genetic studies to study pathogenetic mechanisms of the disease.

Ep syphilis expert panel of control materials for serological diagnostics of syphilis

Abstract

GNTSD Rosmedtekhnologiy (State Research Center for Dermatology and Venereology of the Federal Agency for High-Technology Medical Aid) created a set of agents entitled "EP SYPHILIS expert panel of reference serums for serological diagnostics of syphilis" intended for internal laboratory and external quality control for these types of clinical laboratory tests. The expert panel includes four samples of reference materials: two samples containing the anti-T. pallidum antibody in high and low concentrations, and two other samples without the antibodies. According to the international recommendations for production of biological standards and reference samples, EP SYPHILIS control materials are prepared in a native liquid form ready for administration; the materials are produced based on the human blood serum matrix. The infection security of the control materials is ensured due to the preliminary examination of raw materials for the presence of markers of hemotransmissive infections (HIV and human B and C hepatitis) and thermal inactivation (within three hours at the temperature of 56 С). The specific activity of these control materials during the warranty period (six months) is ensured by means of adding Proclean 300, a stabilizing agent (preservative), in the final concentration of 0.1%, and sterilizing filtration. Regular application of these control materials in clinical and diagnostics laboratories of health care institutions is aimed at standardization and improvement of the quality of tests intended for diagnostics of syphilis.

Clinical and immunological aspects of recurrent, bacterial and viral skin affections

Abstract

The authors analyzed the subpopulation composition of lymphocytes in the peripheral blood of 69 patients suffering from chronic recurrent furunculosis and herpes at the stage of remission. They revealed similar changes in the form of an increased content of T-helpers and immunoregulatory index. In herpes patients such changes are accompanied with the reduced number of natural killers while in furunculosis patients changes are related to the reduced amount of activated T-lymphocytes. These changes do not depend on the relapse duration or frequency but are related to the number of exacerbations in the course of the disease.

Abstract

The authors describe a clinical study of a patient who was suffering from psoriasis for many years and was diagnosed with pemphigus foliaceus after an additional examination. When the patient was hospitalized, the pathological process in the skin was almost close to erythroderma. Systemic and external therapy failed to produce the desired effect. The diagnosis of pemphigus was confirmed by the results of cytological and histological examinations. The authors describe problems with diagnostics and treatment of the diseases.

Combined method of treatment of palmar and plantar warts

Abstract

The article presents the results of an open-label comparative randomized clinical study of a population of outpatients of FGU UrNIIDViI (Ural Research Institute for Dermatology, Venereology and Immunopathology). The study involved 60 patients with viral palmar and planar warts. The subjects were divided into two groups. Thirty patients from the main group received an immune-modulating drug for 10 days as a therapy with the simultaneous removal of warts by the cryodestruction method with the use of liquid nitrogen, and thirty patients from the control group received treatment for their warts only by the method of cryodestruction with the use of liquid nitrogen. Introduction of an immune-modulating and antiviral drug in the complex therapy of patients with viral palmar and planar warts in a combination with cryodestruction had a clinical effect in 93.3% of patients.

Skin photoaging: current aspects

Abstract

The author provides current data about mechanisms of the damaging effect of ultraviolet rays on our skin as well as describes constitutional skin phototypes (based on the Fitzpatrick scale) and clinical stages of skin photoaging (based on the Glogau classification system). The author assessed the extent of changes in the cell structure at different epithelial tumors of skin developing against the background of progressing skin photoaging. The study used molecular markers showing the extent of proliferative activity and differentiation of keratinocytes (anti-Ki67 antibodies, PCNA, P53, Bcl-2, matrix metalloproteinases MMP-1, -9 and their endogenous inhibitors Т1МР-1 ,-2) as their interpretation helps understand the processes of induction and promotion of oncogenesis induced by ultraviolet rays. The author also describes methods of photoaging prevention and treatment.

Abstract

The author describes the results of administering an injection drug, a biorepair product belonging to the HYALREPAIR product line from the class of non-medicinal macromolecular therapeutic products (Registration Certificate No. FSR 2010/06572) for correction of involutive skin changes. Based on changes in such indices as elastometry, skin fold thickness and value of the cervicomental angle prior to and after a course of treatment, the author made a conclusion about high efficacy of the drug not only for facial skin rejuvenation procedures but also for correction of excessive fat deposits and ptosis in the bottom third part of the face.
